For Business Hosting plans and above, yes, this type of service is termed Add-On Domain, you can indicate which folder you want the web pages to be served from. It can be different from your main website. You can also have it redirected to one of the web pages of your main site.

Can I point all my domains to the same main website?
Yes, this type of service is termed Domain Parking, it is unable to choose the folder from which the web pages should be served from. By default it is set to your main website.
